In WinXP the latest Defraggler 20.201, my backup drive contains large files of 1,528,000KB. It has been running now for about 12 hours and still has 10 fragmented files, 20% fragmentation. Apparently it takes at least an hour to defrag each file.
Is there a file size limit that would cause this to be faster, or another setting, or anything that can be done to speed up this operation.

Wow 12 hours that's a long time. Have you tried using Windows Defragmenter? Or maybe there's a missing file that could be causing this slowness in Defragler when it was installed.
Defraggler works great on normal smaller files which I have up to perhaps 500MB. It's simply these larger compressed Acronis Image files that are 1.5GB. Perhaps the program has difficulty with the compression?
I can't answer your question, but if those Acronis files are important backups, I would most definitely "Verify" their integrity after defragging them.
For this reason I avoid defragging my Macrium Image backups, and if they become fragmented I would rather make a new one than defrag and verify.

Thanks, and I agree 100% and I'll verify today before putting the external hard drive in another location.
