I use Windows 7 64 bit and CCleaner does not clean the username/appdata/local/temp files however if you start in safe mode and run CCleaner it seems to clean this folder.
Can this be at least a option with a reboot required ?

I doubt that CCleaner is more aggressive in SAFE mode.
In non-safe normal mode amongst the many extra processes and applications that are running,
perhaps one of them has got a lock on one of its TEMP files.
I suggest you identify what has put the "dirt" into username/appdata/local/temp
Also make sure you have the option to clean temp files younger than 24 hours selected
