Usually it's not something that can be blamed on Defraggler. But for troubleshooting here's some inspiration:
1. Are you running the latest version v1.14.159?
2. Are you sure you don't have malware? You'll need to scan with 3 or 4 different programs in order to make sure. Apart form the main security programs we all have extra scanners (non-realtime) installed with the ability to remove findings. Visiting online scanners frequently is also advisable.
3. How often do you run chkdsk/scandisk and scannow? Files do get damaged from ordinary use. Often times problems like the one you have can be solved by running these procedures.
4. A visit to Microsoft's free scanner listed in my signature below would probably also be of benefit. It's no ordinary scanner.
5. And a long shot: keeping drivers updated by checking the list on the manufacturer's site have solved many a problem.
6. Another long shot: check if you need updates such as Microsoft Visual C++. The freeware program Secunia PSI can do that for you. Microsoft Update as well but for fewer programs. Use the Custom button in there. Not Express. Secunia PSI: http://secunia.com/vulnerability_scanning/personal/
