When I'm booted in Win 7, and open Defraggler, at the top it shows which drives are available for defragging, including the Mac side "Macintosh HD" (E:). Would it be safe to use Defraggler on that...?

Post-2011 iMac's use Solid State Drives. Do not defrag a solid state drive.
