Earlier i had run the de-fragmentation tool (Piriform, Defraggler) on all my drives in my Laptop. From then onwards the audio in my laptop is not working. I generally get a red-crossed symbol in my audio speakers button on the right hand task bar of my laptop. Occasionally at times the audio is working. I am not being able to figure out what might be the reason for the malfunction of my audio. Please suggest me some solutions if any in this regard.
Try disabling then re-enabling your audio device, or reinstalling the drivers for your sound card. Seems odd that a defrag that finished without problems would cause such an issue.
Try:
- Adjust audio slider (sometimes machine is muted, & this unmutes it).
- Update BIOS from manufacturer website (if your are experienced at things like this, as they contain bug fixes). Turn off your A/V till reboot so it won't block it.
- Use default OEM audio driver from manufacturer website. Sometimes, default may work better.
- System Restore -> Sometimes, this can fix or repair problems.

* Had a friend once, who had ME on their machine. Blue-screened on XP update, till they updated the BIOS. Then, it worked for years with no problems. Newer BIOS may work better with your audio.

so let's start with OS version? Are you running windows 7 or 8? Or an earlier one?
Try checking whether volume is MUTED or not. Some programs may change it without you noticing.
I am using windows vista Home premium
please open computer management (right click computer on either the desktop or the start menu choose manage)
go to services (the first heading under services and applications)
find the service called "windows audio service" is it marked as automatic and started?
also if the red x is there but audio is happening, I've had that in both my vista and 7 do that. I think the icon just isn't updating after the service starts. sually mute and minute will change the icon.
